Globelink supports B/L or booking numbers. A number of any other type will not resolve for this carrier, whichever tool you submit it from.
Globelink is identified by the carrier code YSGS - its SCAC. It is what the API takes as its carrierCode parameter, and what identifies the line on a bill of lading.
Yes - one REST call, documented in the API reference. The same balance covers the API and the web app, so there is no separate fee for API access.
